Hazards to Domestic Animals
Simparica TRIO is for oral use in dogs only; do not administer to cats or other species. This variant is not appropriate for dogs below 44.1 lbs (20.1 kg) or younger than 8 weeks of age. Sarolaner is an isoxazoline-class compound linked to neurologic adverse reactions including tremors, ataxia, and seizures; use with caution in dogs with a history of seizures or neurological disorders. Zoetis has not evaluated the safety of Simparica TRIO in breeding, pregnant, or lactating dogs; a veterinarian should be consulted before use in these animals. Debilitated dogs or those on concurrent medications require veterinary assessment prior to administration.
Possible Side Effects
Monitor your dog after each Simparica TRIO administration, with close observation during the 24 to 48 hours following the initial dose.
The adverse reactions below were captured across three field safety studies conducted by Zoetis; primary incidence data are drawn from 272 dogs dosed monthly for up to 11 treatment cycles:
- Vomiting: Reported in 14.3% of treated dogs in the primary field study; the highest-frequency adverse reaction across all three studies, also observed in dogs enrolled in the flea and roundworm treatment studies.
- Diarrhea: Recorded in 13.2% of treated dogs, with or without blood in post-approval reports; incidence was higher in dogs that harboured active roundworm infections at the time of first administration.
- Lethargy: Observed in 8.5% of treated dogs; generally self-limiting and transient across the study population.
- Anorexia: Reported in 5.1% of treated dogs, at a rate comparable to the active control group in the primary study.
- Polyuria: Observed in 3.7% of treated dogs within the primary field study.
- Hyperactivity: Reported in 2.2% of the Simparica TRIO group versus 0.7% in the active control group.
- Seizures: Confirmed in one dog in a controlled laboratory study 16 days post-administration; post-approval FDA/CVM data classify seizure as a known risk across the isoxazoline drug class.
- Muscle tremors and ataxia: Present in post-approval adverse drug experience reports filed with FDA/CVM; frequency cannot be reliably quantified from spontaneous post-market submissions alone.
