Description
Simparica TRIO (sarolaner, moxidectin, and pyrantel) is a triple-active oral chewable manufactured by Zoetis, delivering 6 mg sarolaner, 0.12 mg moxidectin, and 25 mg pyrantel per tablet, calibrated for small-to-medium dogs stepping up from the puppy weight band into early adulthood. Each monthly dose addresses fleas, six tick species, heartworm larvae, tapeworm prevention, and intestinal nematodes including roundworms and hookworms.
Recommended for:
Dogs weighing 5.6 to 11 lbs (2.6 to 5 kg) from 8 weeks of age and older.
Indication/Coverage
For dogs suffering from, or at risk of, mixed parasitic infections, Simparica TRIO addresses the following conditions:
- Heartworm prevention: Prevention of heartworm disease caused by Dirofilaria immitis through elimination of susceptible L3/L4 larval stages.
- Flea treatment and prevention: Elimination of adult fleas (Ctenocephalides felis); prevents re-establishment of flea infestations on a monthly cycle.
- Tapeworm prevention: Prevents Dipylidium caninum infections as a direct consequence of eliminating Ctenocephalides felis vector fleas on the treated dog.
- Tick treatment and control: Treatment and control of infestations with Amblyomma americanum (lone star tick), Amblyomma maculatum (Gulf Coast tick), Dermacentor variabilis (American dog tick), Ixodes scapularis (black-legged tick), Rhipicephalus sanguineus (brown dog tick), and Haemaphysalis longicornis (Asian longhorned tick).
- Lyme disease prevention: Prevents Borrelia burgdorferi infection as a direct result of eliminating Ixodes scapularis vector ticks on the treated dog.
- Roundworm treatment and control: Treatment and control of Toxocara canis (immature adults and adults) and Toxascaris leonina (adults).
- Hookworm treatment and control: Treatment and control of Ancylostoma caninum (L4 larvae, immature adults, and adults) and Uncinaria stenocephala (adults).
Key Benefits
The features below reflect what the Zoetis product label and supporting clinical data established for dogs in the 5.6 to 11 lb weight band:
- Broad-spectrum single dose: One monthly tablet addresses ectoparasites, heartworm larvae, and intestinal nematodes simultaneously, eliminating the need to coordinate separate administration schedules across multiple products.
- Eligible from 8 weeks: Dogs as young as 8 weeks and as light as 5.6 lbs (2.6 kg) qualify for this variant, covering the window when parasite exposure risk is already present but body weight has outgrown the starter band.
- Rapid flea elimination: Simparica TRIO begins killing fleas at 4 hours post-administration and reaches 100% efficacy at 8 hours, reducing the probability of flea egg deposition within the first day of dosing.
- Sustained tick efficacy: Against I. scapularis, the product reduces live tick counts by at least 94.2% within 24 hours of infestation and maintains efficacy through 28 days, covering the full monthly dosing interval.
- Multi-stage nematode clearance: Pyrantel pamoate acts as a depolarizing neuromuscular blocker targeting hookworm across three life stages (L4, immature adult, adult) and roundworm at immature adult and adult stages, clearing established intestinal infections with a single dose.
- Lyme disease interruption: By eliminating I. scapularis vector ticks, Simparica TRIO was demonstrated in two controlled laboratory studies to prevent Borrelia burgdorferi transmission at 28 days post-treatment, directly relevant to dogs in tick-endemic regions.
- Oral format for topical-resistant dogs: For dogs in the small-to-medium weight range that resist spot-on application, the flavored chewable format achieves full systemic distribution without requiring physical handling of the coat or skin contact.
- Flea reproductive cycle disruption: In a controlled study, Simparica TRIO killed fleas before egg deposition could occur for 35 consecutive days, limiting environmental flea burden alongside on-animal treatment.
Caution
Hazards to Humans
Keep out of reach of children. This product is formulated for oral administration to dogs; human ingestion requires immediate medical attention. Wash hands after handling tablets and before food contact. Avoid transferring tablet fragments to eyes or broken skin.
Hazards to Domestic Animals
Simparica TRIO is for oral use in dogs only; do not administer to cats or other species. This variant is not appropriate for dogs below 5.6 lbs (2.6 kg) or younger than 8 weeks. Sarolaner belongs to the isoxazoline class, associated with neurologic adverse reactions including tremors, ataxia, and seizures; use with caution in dogs with a history of seizures or neurological disorders. The safety of Simparica TRIO has not been evaluated in breeding, pregnant, or lactating dogs; consult a veterinarian before use in these animals. Dogs that are debilitated or receiving concurrent medications should be assessed by a veterinarian prior to administration.
Possible Side Effects
Monitor your dog after each administration of Simparica TRIO, with particular attention during the first 24 to 48 hours following the initial dose.
The adverse reactions below were recorded across three field safety studies conducted by Zoetis, with primary frequency data drawn from 272 dogs treated monthly for up to 11 cycles:
- Vomiting: Reported in 14.3% of treated dogs in the primary field study; the most frequently observed reaction across all three studies, including those focused on flea and roundworm populations.
- Diarrhea: Recorded in 13.2% of treated dogs, with or without blood noted in post-approval reporting; occurrence was elevated in dogs with concurrent roundworm infections at treatment initiation.
- Lethargy: Observed in 8.5% of treated dogs; generally transient in nature.
- Anorexia: Reported in 5.1% of treated dogs, at a rate comparable to the active control group.
- Polyuria: Observed in 3.7% of treated dogs in the primary field study.
- Hyperactivity: Reported in 2.2% of the Simparica TRIO group versus 0.7% in the active control group.
- Seizures: Documented in one dog in a well-controlled laboratory study 16 days post-administration; post-approval FDA/CVM adverse event data list seizure as a known risk across the isoxazoline class.
- Muscle tremors and ataxia: Included in post-approval adverse drug experience reports; frequency cannot be reliably estimated from spontaneous reporting data alone.
If any of these signs persist or worsen after dosing, discontinue use and contact a veterinarian.
